Ulm and Augsburg are worth a stop but don’t need a full day. Harburg would be nice and not too far. Nördlingen as well. Could make a second day. Nürnberg is worth a full day. So is Munich.
If you’re a car nut, I would add the Auto&Technik Museum in Sinsheim. Not so much a museum as a wild collection of everything with a motor, but you’re driving right past it.
For the part directly south of Frankfurt (A65 & A67) up until Heidelberg, there are pretty scenic routes both to the East and the West of it. I would recommend going West of it, on the other side of the Rhine, you can drive through Rheinhessen and down the „Deutsche Weinstraße“ for example, then cross the Rhein again between Mannheim and Karlsruhe to get back to your originally planned route.
